Description:This laboratory manual is the outcome of teaching in this area for the past 12 years to our M.Sc. Biotechnology students under National Biotechnology Training Program of DBT, New Delhi. This book consists of ~30 Chapters and is organized into 5 Parts. Part- I essentially discusses the General Instructions for using the laboratory and Good Laboratory Practices. Part- II presents in detail the common Biochemical Techniques used at B.Sc. and M.Sc. levels, whereas Part- III, deals with Separation Techniques such as Electrophoretic Techniques (SDS, Agarose, IEF, 2D gels, Pulse-Field Gels and Capillary Electrophoreses) Basic Chromatographic Techniques (Adsorption, Paper, TLC, IEC, Gel filtration, etc., and also Advanced Chromatographic Techniques such as Hydroxyapatite, DNA-Cellulose, dHPLC, UPLC, FPLC); MALDI-ToF (Tandem MS) are also described. Part- IV deals with Enzymes and Enzyme Techniques such as basic enzyme kinetics, assay of enzymes such as amylases, alkaline/acid phosphatases, invertases, proteases, DNA polymerases, Restriction endonucleases, purification of enzymes including restriction endonucleases and zymogram analysis of enzymes. An Appendix (Part- IV) includes all the reference data of various experiments. In each Chapter the background and basic principles of the experiments are explained in sufficient detail with illustrations and along with the experimental protocols. Experimental results (wherever available) obtained under Indian laboratory conditions are given for students' reference. Almost all these experiments were done by the author himself here and repeated many a time by the M.Sc. students over these years. The author sincerely hopes that this book will be useful to B.Sc., M.Sc., M. Phil. and Research students of Biotechnology and other areas of Life Sciences.
