Very briefly, the program is a sequence of transcribable DNA sequences that regulate the expression of protein-coding genes in each generation of the cell clone through non-coding RNA transcripts (called nuclear messenger RNAs, nmRNAs, which include all the many classes of non-coding RNAs that have been observed). The MDP is proposed to evolve through terminal additions and to be located in the pericentromeric heterochromatin (alpha satellite) DNA. The manuscript summarizes how I came to concoct the hypothesis and provides a conventional review of the current state of our understanding of differentiation and reprogramming of cells in complex, multi-cellular organisms. I have tried to make the general ideas understandable by people with a general interest in biology and provide enough substantive information to make it worthwhile reading for researchers engaged in evolutionary developmental biology (EVO-DEVO) and genome regulation.
